Prolonged Fasting With Capivasertib (Truqap®) for Metastatic Breast Cancer

Fast-Her: Fasting Effects on Breast Cancer Treatment

Purpose: To study the safety, effects (good and bad), and anti-cancer activity of whether prolonged fasting is acceptable and manageable for people with metastatic breast cancer who are taking capivasertib (Truqap®).

Who is this for?: People with metastatic (stage IV) breast cancer who are postmenopausal or are taking medicine to suppress ovarian function. You must currently be taking capivasertib (Truqap®) and have been on a stable dose for at least 2 weeks. You must also have a body mass index (BMI) of 25 or higher, wear a continuous glucose monitor, and track your meals with a mobile app. This trial is available at the University of Minnesota in Minneapolis, MN. What's involved?

<p class="seamTextPara"> You will be assigned to 1 of 2 groups: </p> <p class="seamTextPara"> <i class="seamTextEmphasis">Group 1</i> </p> <ul class="seamTextUnorderedList"> <li class="seamTextUnorderedListItem">A normal eating period, then a washout period (a break to let the effects of the first plan wear off), then a prolonged fasting period.</li> </ul> <p class="seamTextPara"> <i class="seamTextEmphasis">Group 2</i> </p> <ul class="seamTextUnorderedList"> <li class="seamTextUnorderedListItem">A prolonged fasting period, then a washout period, then a normal eating period.</li> </ul> <p class="seamTextPara"> <i class="seamTextEmphasis">During the prolonged fasting period, you eat one meal a day (dinner). Water, black coffee, and tea are allowed during fasting hours. During the normal eating period, you follow your usual eating pattern.</i></p>

What's being studied?

<ul class="seamTextUnorderedList"> <li class="seamTextUnorderedListItem">This study is testing whether going without food for longer stretches (prolonged fasting) can be tolerated by people with metastatic breast cancer and whether it can help their cancer medicine work better. </li> <li class="seamTextUnorderedListItem">Capivasertib (Truqap®) blocks a signaling pathway (the PI3K pathway) that breast cancer cells use to grow and survive. Researchers believe a fasting state may boost the anticancer effects of this medicine. </li> <li class="seamTextUnorderedListItem">Because this is a crossover study, each person tries both a normal eating plan and a prolonged fasting plan so the two can be compared. </li> <li class="seamTextUnorderedListItem">Researchers will look at blood samples, laboratory and cell studies, and patient characteristics to understand how, and for whom, fasting might improve the effect of capivasertib.</li></ul>
